چکیده

Overview Purpose: To develop fast and sensitive approaches for the analysis of underivatized free amino acids and peptides using the latest UHPLC column technology and charged aerosol detection. Methods: Presented here are two distinct methods using the same UHPLC analytical column. The first method uses large amounts of ion-pairing agents to resolve 18 free amino acids. The second method uses less ion-pairing agent and an inverse gradient approach that enables a direct comparison of charged aerosol detection to traditional UV techniques. Protein digestion is monitored on the autosampler tray. Results: The first method enabled the analysis of 18 amino acids in under 9 minutes with low nanogram sensitivity for all analytes. The tryptic digest method not only allowed the separation of amino acid residues and peptides, but the time course of digestion could be followed using this automated system. The results showed that charged aerosol detection is a complementary technique to the well established low wavelength UV with added sensitivity for small peptides and many amino acids.

Fast UHPLC Methods for Analysis of Amino Acids

Overview Purpose: Methods that offer both rapid and sensitive measurement of amino acids are essential to neurochemical research. The ability to decrease analysis time enables an improvement in temporal resolution when studying amino acid neurotransmission. Methods: Two methods using pre-column derivatization were developed: first, a fast isocratic method with electrochemical detection for the ...

Analysis of underivatized amino acids by capillary electrophoresis using constant potential amperometric detection.

A mixture of native (underivatized) amino acids is separated by capillary electrophoresis under alkaline conditions (pH approximately 12) and amperometrically detected with a copper-disk microelectrode. A simple design facilitates capillary-electrode alignment without the need for micropositioning equipment. The limits of detection for the amino acids are in the low microM concentration range, ...

Ultrasonic acoustic wave detection of single or capillary electrophoretically resolved underivatized amino acids.

Acoustic wave detection of various underivatized amino acids that were hydrodynamically introduced into and electrokinetically migrated along a capillary tube has been achieved. The detection principle is based on the measurement of the ultrasonic absorption and density change of the aqueous amino acid samples as they pass by the detection zone. Acoustic wave detection of underivatized leucine ...

Performance of 2-Amino Tetraphenyl Porphyrin as Stationary Phase in RP-HPLC of Amino Acids

The search for new stationary phases has been one of the predominant concerns in high performance liquid chromatography (HPLC) in order to achieve better resolutions, longer column lives, and reduce the time of analysis. A chromatographic packing for separation of underivatized amino acids (AAs) were prepared by dynamically coating 2-amino tetraphenyl prophyrin (atpp) on a C-18 reversed-pha...
